1) 0.5ml의 sample을 15ml conical tube에 넣어준다.
2) 15ml conical tube에 0.5ml 100% ethanol을 더해준다.
3) 15ml conical tube에 들어있는 혼합물을 vortex 해준다.
<n-Hexane>
4) 15ml conical tube에 2.5ml n-Hexane을 더해준다.
5) 15ml conical tube의 혼합물이 잘 섞이도록 vortex 해준다 (1분)
6) 만들어진 sample 을 3000RPM 에서 5분간 원심분리 해준다.
7) n-Hexane 층에서 1.5ml 만큼 따서 새로운 15ml conical tube에 옮겨준다 (n-Hexane 층은 가장 윗층)
8) flowhood 에서 sample을 nitrogen purge 해준다 (30℃)
9) purge가 끝난 샘플에 150㎕ isopropanol(2-propanol) 만큼 넣고 dissolve 시켜준다.
(프로토콜에 따라서는 500㎕ 만큼 넣어 줄 수 도 있다, 하지만 이 때 dilution factor 가 바뀜을 명시해야한다)

10) 0.45㎛ syringe driven filter unit을 이용하여 sample을 HPLC vial로 옮겨준다.
11) HPLC 분석
